Port Lions has no overland access roads; visitors must travel by land or water. Several local commercial air carriers service Port Lions, with daily mail flights scheduled and charter flights available as well. Air access to Port Lions is always weather dependent. Port Lions also receives regular commercial ferry service, with the State Ferry visiting bimonthly between May and October.

The road system in Port Lions is more extensive than in most other Kodiak villages. The roads are generally in good condition, with many privately owned vehicles in the community, as well as ATVs and some larger trucks.
